Se habla espanol? means "Is Spanish spoken?
Se hablaen espanol has either a typo, it's misspelled, or it's incorrect. Se habla en espanol is probably what was meant and is grammatically wrong. Se hablan espanol could also be what is meant. In any event, the meanings would be as follows:

Se habla espanol? Is Spanish spoken? (Does one speak Spanish?)
Se habla espanol. Spanish is spoken here. (One speaks Spanish.)

Se hablan espanol: They speak speak Spanish. (This is incorrect Spanish if this is the intended meaning. I should be Hablan espanol.)

Se habla en espanol. (It might be an attempt to say that someone is speaking in, or speaks, in Spanish. But it is very incorrect.)

Se habla español? = Is Spanish spoken?

that in case you are asking if spanish is spoken in that place, but on a sign of a shop if you see

Se habla español = It means Spanish spoken here. In that case it has no question mark. It is a statement.

literally: Is Spanish spoken?
However the ”in” strikes me as strange!

When the sentence finishes with a question mark, it means:
Do you speak spanish? (talking about a place)

But when it finishes with a period, means:

Here people speak spanish.
