It depends on what pill you are taking. Some have immediate effect and others have to build up a certain level in your blood before they can take effect (which could take days for certain meds).
To make it work "faster" and assuming the pill takes effect once it is absorbed from your GI tract you can do a couple things:
Take it on a nearly empty stomach (assuming it does not upset it and it isn't an NSAID) or stay away from fatty meals because they slow down digestion.
Lie on your right side (the stomach empties towards the right).
I disagree with someone's answer of being active afterward. The blood flow to your GI tract increases when you take a pill, as does the the rate of absorption. If you are active, the blood flow moves away from your GI tract and to your muscular system (arms and legs). This results in a slower rate of digestion, and a slower effect of the medication to take place.

It depends on what type of pill it is. For example, if it's antibiotics you may have to take them for a few days before you start to feel better. If it is a hormone, such as a birth control pill, it may not be effective for several weeks. Analgesics may help in 15 to 20 minutes.

It depends on what kind of medication it is...what kind of capsule...etc. Best to follow the direction on the pill bottle to make it work the fastest. And talk to your doc if you are really want to know more.
