Radioactive spider? You've been watching too much Spiderman. Anyhow, radiation, like any other mutagen, has the potential to damage DNA. The location within the DNA that is damaged may be benign, latent (one of multiple mutations required) or, initiate cancer. If the DNA damage is not repaired by the cells natural repair mechanism or the mutated cell is not destroyed by either suicide (apoptosis) or the bodies immune system, cancer may propagate.

As for cancer, a short answer is cancer is the uncontrolled, rapid growth of abnormal (immature or not fully differentiated) cells.

cells have a few rules that they must live by

they are surface dependant-they will not grow unless they are anchored to a surface
they will stop reproducing when they feel touch on all sides-they will not grow on top of each other

radiation damages cells dna this could cause a cell to form differently than neighbor cells and rapidly divide. Cells have a self destruct program they use when thier natural inhibitions are not working correctly, and other cells can kill themselves to stop the spread of rogue cells. Although, if these safety measure do not work then you will begin to form turmors, and thus you have cancer.

They send out radiation, and that's how those things cause cell damage and therefore they cause cancer.
