It takes practise. I guess you are talking about getting confidence when faced with a particular situation, such a public talking. First of all know your subject. Second, know what you audience expects from you (even if this is just one person you are talking to). Knowledge build confidence.
Finally, if you are still nervous, imagine your audience naked and how ridiculous they look.
After all, they are only people; just like you.

Depends on what you are nervous about.
-If you are talking about anxiety, try some biofeedback techniques like breathing, and temperature focus control. You may also see your doctor to make sure it's not another condition like thyroid disorder.
-If it's in anticipation of performance, like testing or a speech, do a really good job studying, then stop 24 hours before and let yourself know you have done all you can. If you train your body to be okay with what you have done, you will start to feel less nervous in anticipation.
-DON'T drink caffeine or consume lots of sugar. It will only exaccerbate the sensation of nervousness. Drink lots of water to keep yourself healthy.
-Don't procrastinate - if you're nervous about completing a task (say, visiting the doctor), just do it! The longer you wait the worse it will be in your mind, but not in reality.

You should try to get into a laid back mindset right before you do something that makes you nervous. Just think as if you were at home and in your PJs and having a chat with someone you know really well.
