The are some portions where the air pressure differentiate and that's when turbulence happen, the clouds for example, because of their configuration they don't have the same pressure at any point of the cloud.

Turbulence is caused usually when something disrupts the normal air flow. Whether it be a thunderstorm, mountains, or even another aircraft.
