It's a question of "did you understand the humor in my last statement," just in fewer words. Unfortunately, people who have to ask all the time aren't that funny.

They're trying to find out if you actually got the punch line, and if your reaction was positive then they know that you understood it.

to make sure the audience understands the joke.. and if not..then they will explain about it.. lol.. also.. it could be used to promote the comedy atmosphere.
