I can't imagine that there would only be "1"..... Since we all have different desires and cultures, it would completely depend on what we like and want to do at a hotel/resort!! Reality is what is number one for you, may not be the number one hotel for me!!
I know that Paradise Island in the bahamas has the most expensive hotel suite (and there is only one) in the world or so they claim.

L'Hotel Meurice in Paris. Absolutely stunning. It's like staying in an 18th century Palace. The restaurant, the Grill room is exquisite and the service impeccable. For the non-french speaking, almost all their staff spoke English.
The price is like for the Yachts, if you ask how much , you can't afford it.
Among their customers, past and present Salvador Dali, King Juan Carlos ,many Arab Princes.(as a matter of fact I think It is own by a Arab corporation.)
