I know that "Que" means what, but I'm not sure if it should be used in the same way or if that would be considered rude.

It's better if you use "perdón" or "mande" instead of "qué".
"Qué" sounds rude in this case.

Qué? is a rude way of answering.
It'd be better if you answered:
-Si? (this can also be followed by a title like señor, señorita, señora, patrón, jefe, joven, etc.)
-Qué desea?
-En qué le puedo ayudar?
-Diga
-Mande

Now, if you didn't catch the question you could ask
Cómo dijo?
Podría repetir la pregunta?
Disculpe, no escuché bien su pregunta.

¿perdon? (like parden me) is polite
¡mande! (is the command to send) very common. used with people you are familiar
with.
¿que? what? impolite,, not recommended.

you can also say "como como?" instead of "que" if you didn't hear what someone said

Depends on where you are. In Mexico it's ¿Mande? and in Spain it's ¿Cómo?

I wouldn´t use Qué. You can never go wrong with ¿Perdón?

It is rude, and lower caste to say "que" when you mean "what". What you say is "Mande?" pronounced "Monday?". Or you can say "?Mande Me?" Like please repeat. I found that one out the hard way, my partner is Mexican. LOL

you can say "perdón, no lo entendí bien"
or "que?", como?; como dijiste? ; they are all fine

I don't know what pardon is in spanish but say that, it is much more polite.
