It's when he's able to accept the responsibilities of being a mature man. That means he will think of others before himself and he'll be respectfull of others and help others when needed, not when ask. For example. If a big girl who no one considers pretty drops her things at school and the guy does not pick it up for her than he is not a man yet. But if the guy was not concern about self image and picks up the girl's things, then I'll consider him a man. He's able to not care about what others think but do what he believes is the right action to take.

Well look for the lump on the throat for one and the big hands should give it away as well ive never seen any woman with really big hands also the obvious would help

he is responsible, has a backbone, will do what's right no matter what, is polite, doesn't feel the need to put others down to make himself look better, puts others first, and doesn't need to be reminded to keep up on his personal hygiene
