Depends on what you mean. Music? Clothing? Subculture?

Music:
Punk is kind of rocky in a sort, the kind of stuff that one ascociates with a skater. Think: Blink 182, Sum 41, Good Charlotte
Emo CAN be depressing, but is mostly what the name says: emotional. Lyrics are emphasized with emotion, and are often at least a little dark. Think: Senses Fail, My Chemical Romance, Hawthorne Heights.
Goth... well, there isn't really GOTH music I suppose. Maybe you mean clothes...[there is gothic death metal?]

As far as clothing, punk is bright, tends to encorporate skater tees and maybe skater shoes, and a sort of "fake rebel" kind of look.
Emo for guys involves bangs, chick pants, and band t-shirts.
For girls it involves clothes with gothic themes [like skulls] made someone cute, or overstylized, often in a pattern, such as a shirt with repeated lightning bolts, hearts and stars. Bows in the hair can be incorporated. Emo for girls is often more cute.
The true definition of goth clothing is just BLACK. That is it. Gothic clothing varies a lot, and the only true thing is that goth IS black. The clothing is often dark, not only in color, but in feeling.

As far as subculture, I see that punks tend to hang out more with skater-types.
Emos act like preps even though they were odd clothes. Some act overly depressed, but this is usually just a stereotype.
Goths are usually darker then emos REALLY are, they can be morbid, but aren't always depressed.

The difference is mostly based on the music they listen to,and by listening to a certain kind of music,you become part of that culture.
*Punk: some REAL punk bands would be the sex pistols,ramones,the clash etc,punk music usually tends to be somewhat political, atacking the unjustice practiced by the government.The orientation is Anarchy, for which they fight against nazis,comunists,laws and social layering.As for the clothing, punk doesn't have an uniform, punks go with the ideea of do it yourself so it is mostly about improvisation.A lot of people asociate punk with black but as Johny Rotten said punk is more like"neon green",it is about being what you are and what you feel.
*I don't know much about goths but they listen to gothic music like cradle of filth,children of bodom etc.They are more atracted to dark things as in dark colors and mythical creatures, witches and stuff like that.
*And emo...it's kind of new but I guess it's quickly becoming a culture.Emo stands for emotional and it's about people with deep and powerful feelings but they tend to take it to far if you ask me...One of the music genres they liten to is emo-punk like panic at the disco and fall out boy.And I know they listen to AFI,but as far as I'm concerned that doesn't even come close to anything punk...
*I don't know so much abot goths and emos but you could look it up on www.wikipedia.com you can find a lot o stuff about almost any musical culture.

A Punk is into anarchy, rebellion, and non-conformity. They listen to angry, loud music, and wear scraps of clothing; mostly found in thrift stores or dumpsters. They are very into D.I.Y.

Punks follow with the idea that life is to be lived, and to the most extreme form of living. They love to roller blade, bungee jump, sky dive, and anything that spells danger, because they want to push the boundries of society.

A Goth is into the dark side of life. They find beauty in ugliness, love romance and desire, as well as death and decay. They listen to darker sounding music, classical and opera, and modern music about dying and loving.

They wear Victorian/Edwardian clothes, or Industrial/Cyberetic type clothes, not always black. Goth's love to be creative, poetry, acting, music, painting or drawing, and they adore D.I.Y.

An Emo is a fairly new subculture. It's about conformity and following trends. Emo's are young children who have entered Puberty and are learning about identity, self-image, and what they want out of life.

Emo's listen to watered down Punk and Indie type music, and wear whatever is the latest trend. It used to be pull-over sweaters, now it's dinosuar hoodies. They are not into the D.I.Y culture, because they can buy something that looks D.I.Y.

Real punks died out in the mid-80's. All of these new kids are just spoiled brats.

Emo is the single most repulsive thing in the universe. Why, oh why did God allow such a thing in this world?

Gothic is what rich kids think vampires are supposed to be. Truth is, I'm pretty sure that if I were a vampire I'd be pretty ticked off with the way these kids these days are ruining my good name.

This is how I see it
Gothics wear trench coats, listen to music where you can't hear the lyrics through the screaming (like cradle of filth).
Punks are like skaters they pull their socks up and listen to teenager music like Blink 182.
Emos are kind of a bit of both, They wear skater type clothes but they are black. They listen to music like AFI and the guys wear girls jeans.

yea someone please explain to me what the f*** emo is...grrrr if it is what i think it is ..whoever invented it and its name are so ..uhh..stupid does not do it ..i need a better word... i hate it its so teenagerish haaha so stupid and unoriginal it all sounds lame and like someone is whining...ahhhhhh its the bane of my existence!!!! emo? is it like greenday where every song sounds the same? ahaha with the same nah nah nah nah stupid crap...ahhhhhhhhhh emo!!! why have you done this to me?! dude d - you are so right emo is gay! oh no wait that is rude and not nice to gay people...hmmm...
