You can't feel your spleen in a way that gives you localizable pain information. You are guessing that your spleen hurts because of where you feel pain, but I don't even know if you are correct in localizing your spleen! All I can tell from what you say is that you have pain... somewhere... and you're pretty sure your spleen is in that spot. If you have pain in a specific location, one thing that you can say for sure is that the pain is being transmitted to your brain by way of myelinated nerves. These are the type of nerves that come to the central nervous system from the skin and the body wall, and not from internal organs.
When I ask a patient about the location and quality of their pain, and when I find out that it has focalized to a particular place on the body wall, I can then get some actual localizing information about the internal process. When people tend to be vague about the location of pain, and point to a whole region along the midline of the body, I conclude that the pain is "visceral pain" meaning that it is originating within an internal organ.
In response to your more abstract question, though, yes the spleen can swell, but I am unaware of this being related to any medication. There are specific disease states in which the spleen can become enlarged (splenomegaly) and sometimes when it is too big, it can also accidently sequester away certain kinds of blood cells which would be better off circulating around. This condition is called hypersplenism. Splenomegaly is enlargement of the spleen, while hypersplenism is the functional problem of the spleen filtering out too many platelets, leading to thrombocytopenia. In some illnesses, there can be swelling of the spleen as well, such as mononucleosis.

The spleen is without sensory nerves and your spleen can't 'hurt' other structures in the area may do so, but not the spleen. However splenomegaly (increase in the size of the spleen) will cause some discomfort but usually it is not noticed. The antibiotics puzzle me...what disease are you being treated for? For any physician, we need all of the information to make decisions and you have given almost none. We need the present history of this illness, the symptoms, all blood work, X-rays, and the course of the illness. So to give you a definitive opinion as to the cause is impossible. Antibiotics seldom cause any discomfort other than diarrhea, stomache upset or allergic response. Many physicians are guilty of not explaining a health problem clearly, but patients are also guilty at grabbing words rather than understanding the whole idea. Many diseases can cause small to moderate enlargement of the spleen i.e. Mononucleosis, but the only danger is that a fall or other injury may rupture the spleen (which is a recycling organ for hemoglobin and erythrocytes) and such a rupture would be very serious. Please understand your physician or demand a clearer explanation. A second opinion may be needed but you have to know what the problem is first.

The spleen governs your quality of thought. My thoughts have been kind of out of sort lately. Maybe if I surrounded myself with yellow and brown, I'd have a much healthier relationship with my spleen. Maybe I need to start smiling at my spleen. I'm sure it would like that.

I think you should... I know someone who had a small tear in their spleen and all it did was hurt slightly and they almost had to get it removed before someone actually caught it.
