The easiest way is to take the airport shuttle bus. When you come out of customs go to the shuttle desk where they will book it for you and give you a number. You might have to wait 15mins or so but you will be dropped off at your hotel. It costs about $25 plus tip. Look on the main web page for JFK airport for advice.

If you are going to take a taxi, make sure you get on the taxi line for the yellow cabs! There will be a dispatcher who will give you a sheet of information about fares, etc. There is a flat fare into Manhattan of $45, plus tolls and tip.

For the subway, take the AirTran to the "A" train to 42nd Street. You can either walk to the hotel from there, or take a taxi. The hotel is between 8th Avenue and Broadway, and the A train will leave you at 8th Avenue, so it is very close.

There is a good bus service:

This will drop you at either Port Authority (West 42nd Street) or Grand Central (East 42nd Street). From either location you can take a taxi or just walk to the hotel. The Port Authority stop will be much closer for you. The fare is $14 each way.

I've stayed on 51st Street. There will be an airport shuttle bus. Allow half hour to pick you up and an hour to get you there (although could differ depending on number of passengers and route). I can't remember how much I paid but it was about $20 last December.

I warn you strongly against the taxi scam artists who operate at JFK. I got burned by one a couple of years ago.

Take your time, and don't dive into the first taxi you see.
