A poke is a push or a quick thrust. Since you set the scene between a gal and her male friend, I suppose "poking between the legs" might refer to copulation. Bear in mind that this form of expression is quite vulgar and, in my opinion, slightly demeaning to women.

The common phrase is actually "poke you between the eyes" and it basically means that you snap someone back to reality or bring them out of their comfort zone.
