Here's a good site that explaines the "science" behind addiction. Too long to copy and paste the whole article here.

My definition of an addiction is something really simple. Its something you cant stop. And its hard to let go because unlike what most people think, its not just in our head but in our body as well. The cells of our body are also addicted to things, or peptides to be exact. Watch the movie, What the bleep do we know, this explains everything.

We are afraid of change and reality. What matters the most to you, family or school, work, focus on that and work on that alone and know that addiction sets us so far back. I have been there and it's been almost a year since I haven't been addicted to anything. I focused on my son and husband and knew that I was being selfish. It's all mental, it's within us to change, counseling and everything will state the obvious, you have to get control and make your own choices on what you want to do. Good luck, it takes time but it's cureable

some addictions are physical and its your body that is craving the substance and not your "mind" per se. also, think about the positive reinforcement you receive when you give in to temptation, ie. state of euphoria or satisfaction, the endorphin rush you get just from biting into a piece of chocolate cake you've been denying yourself. in many cases the addiction is a combination of psychological, emotional and physical dependence.

If you want a medical answer here it is:
It's because we either develop a physical or psycholoical dependency on an object. That object could be our fallback line when we grow weary and need a place to hide or feel safe in. Your body might just need it in order to operate normally like Ecstacy. Once you've taken too much of it over a period of time, your body grows dependent on it so you aren't depressed all day.

Addition is when you need more of a substance as time goes on.
The original dose does not work anymore
