What is the total number of pilots who are qualified to fly Air Force One? On each flight, how many crew will there be in the cockpit? And how many pilot will be on standby at anyone time?

depends, on commercial aviation, for long haul flights there is usually three up on the flight deck, the first officer and captian who trade turns flying the plane and maintaining the aircraft systems, and the flight engineer who does a further job of maintaining the aircraft systems.

Depends on the length of flight, as with any other airliner. There is a limit to what hours the pilots are allowed to fly without crew rest.

There are a few. You are aware that any Air Force aircraft that the president is flying on is designated Air Force One, It's just not the 747 you normally see him in. When he is flying in the Marine helicopter the designation becomes Marine One!
