The best and cheapest way to get Broadway and Off Broadway tickets is the TKTS booth located in Time Square or in South Street Seaport. How it works is that they have drastically discounted tickets for various shows on sale for that particular day. So only get on line if you have time to watch the show later that day. Also, the booth only takes cash, no credit or check.
Their website is here: http://www.tdf.org/tkts/ It list all the different shows they can potentially have but you won't know for sure until you get to the booth. Also, you have can't choose your seats until you get to the booth either. But you can get really great seats for more than half the orginal price!

My friend, I agree with the first post, however, let me add this little bit of information. The TKTS booth is, at the moment, at a temporary location due to renovation work at Duffy Square, where the booth is normally located. The temporary location is West 48th Street between Broadway and 8th Avenue, on the ground floor of the Marriott Marquee Hotel. Good luck

It depends on how much time you have...There is a 50% discount line int he middle of Times Square where they sell discounted tickets. I have never used this line myself but I have always seen loads of people there.
You can also do directly to the box office on the day and you might get discounted tickets depending on the popularity of the show.
Or you can just use ticketmaster.com to order you tickets, this way you get a seating chart.

times square booth and south street seaport booth. same day as show. if you in the military call USO at port authority for free tickets. saw 3 shows this weekend all free and amazing seats :)
