"Let's" is the contraction for "let us" and does mean the same. Either may be used in conversation. The only difference is in writing. "Let's" would not be used in formal writing such as a report or a business letter, but it could be used in notes, personal letters, etc.

The difference is that let's is a contraction - a shortened form of a word or group of words, with the omitted letters often replaced in written English by an apostrophe, as e'er for ever, isn't for is not, dep't for department. - and therefore one word while let us is two words.
