I can only think of two.... University and cathedral.... maybe an airport for the third.

In the UK all it needs a charter from the royal family.
they usually pick places that have a cathedral however not all do (eg york has a minster instead). Not all cities have an airport though again most have at least a small one. but airports have only been around about a hundred years whereas cities have been around for thousands of years and they can be very small (eg Truro).
I'm sure it is just up to the rulers of the country they are in.

In the UK, it can have been known as a city since time immemorial or have letters patent.
Letters patent are usually given on the basis of size, importance or royal connections.

St Davids City in Wales UK has a cathederal, and a population of around 1.500 people. and thats about all it has, but its a city.
