The Decalogue is recorded both in the Books of Exodus [20:1-17] and Deuteronomy [5:6-21], two of the five Books of Moses (also known as the Torah, Law, or Pentateuch).
The Ten Commandments were given to Moses on two tablets of stone on Mount Sinai.
Also - Jesus gave the Greatest Commanment: Matthew 22:36: "Teacher, which is the greatest commandment in the law?"
37 Jesus said to him, "'You shall love the Lord your God with all your heart, with all your soul, and with all your mind.'
38 This is the first and great commandment.
39 A second likewise is this,'You shall love your neighbor as yourself.'
40 The whole law and the prophets depend on these two commandments."

This depends on what kind of commandment you are trying to find.
The first command to people came just after Adam's and Eve's creation, when God told them to "be fruitful and multiply." (Genesis 1:22) He later told them which trees to eat from and which one to avoid. This is clearly referred to as a command. Genesis 2:16 "And the Lord God commanded the man, saying..."
The first of the Ten Commandments is expressly issued in Exodus 20:3, over 2000 years later, though these commandments would certainly have been known long before that.
THE VERY FIRST commandment mentioned in the Bible may well be that of Genesis 1:3 where God commanded light to exist, and there was light. Alternatively, Genesis 1:9 records God as saying "Let the waters under the heaven be gathered together unto one place, and let the dry land appear." This is later referred to as a command. (see Psalm 33:6-9)

Depends on your meaning of 1st commandment.
Ex. 20 & Deut 5 list the 10 commandments.
but Jesus said Deut ch. 6 contained the greatest.
To Love Jehovah your God with your whole heart, mind, soul, and strength.
