Male cats do it to mark their territory. Cats also do stuff like that when they are angry about something. Not to sound silly but maybe she doesn't like your bro for some reason. Or maybe she is mad at him for something. Has he done anything to take up her personal space somewhere? Cats can be moody it could be any number of things, but it sounds to me like she is upset about something.
