Callanetics is the book for you -- available from most bookshops (or get a video). Doing the exercises makes a visible and palpable difference to your rear end in only a few weeks. If that's your main concern, get the book/video and exercise every day at home. Believe me, it makes a difference.
Going to the gym is a different matter -- it is for building up your stamina and toning you up rather than improving your body shape. Three or four times a week is a good thing, if you can manage that.

1. Squats(hold dumbells if you want)
2. Lunges
3. Running is good for the butt, it made my butt very toned :)
On the treadmill, change the gradient so your running or walking up hill. I used to run using intervals up hill on the teadmill for 20 min, then at the end id change the gradient to the top one and just walk at 5kph taking big steps and clenching my butt on every step. I did that for like 5-10 min. Give it a try.
Or if you prefer go to a step aerobics class. All the stepping is good for the butt. oh and always take the stairs-also good :)
Good luck

Squats...where your butt goes right to the ground...or as close as you can get it
Step ups...where you step up onto a bench while holding a dumbell or barbell
Deadlifts...yeah their not just for your back
