2.1: 2 channel audio with an additional subwoofer channel (3 speakers in total). Typically source material isn't in 2.1 format, it's in 2.0 (or plain old stereo) The low frequencies are sent to the subwoofer by a filter or sound processor in an amplifier

5.1: 5 channel audio with an additional subwoofer channel (typical for surround sound)

http://en.wikipedia.org/wiki/Surround has a whole lot more combinations.

A 2.1 system is
basically a pair of
stereo speakers and
a powered subwoofer.

A 5.1 system is 5 speakers
(2 front, a center channel, and
2 rear surround speakers).

Basically put 2.1 means 2 speakers + 1 subwoofer and 5.1 means 5 speakers + 1 subwoofer. Hope this helps.

Sam's answer is right.
