"Please send your wish to participate to me before 22 November 2006, with an updated CV".
So does he want a motivation letter and a CV? Or just an e-mail with my CV (since sending the CV is expressing a wish to participate)?

Definitely include a letter, even if it's just to say who you are and why you want to participate. Also update your CV and make sure it notes everything that is relevant to the job (or program, education).

Not a motivation letter, just a note saying you want to be a part of it, update your CV, and send the updated version along with the note.

It means send your cv and a letter of acceptance or confirmation, and possibly a letter of personal statement.

I would send both just to be safe, or your cv with a lil attached note expressing your wish.

It's bad English; you can't send a wish. But you're probably better off sending the motivation letter as well.
