In some way your question is a little moot as Greece and Macedon had been fighting against Persia for a very long time before Alexander was born. However, after crossing the Helespont with 42000 men, he won his first major victory against the Persians was the battle of Granicus in 334 BC in which he took control of a large province in Asia Minor

In 334 BC, Alexander crossed over into Asia Minor to begin his conquest of Persia.
