you can tell that a person is lying when:
Facts of some things they have told you before contradict each other.
They get all nervy and anxious when asked about the subject of which a person is lying about and tend to fidget more.
Make no or little eye contact.
Will give such replies like, "Why don't you believe me?" or "You don't believe me? I'm telling the truth" to make them look/sound innocent.
Some may go as far as putting a cup of coffee or pencil case or anything else to sort of create a barrier between you and him/her.
Their voices tend to be high-pitched, tight or tensed.
Liars may tend to go very far into details to make the lie seem like the truth in the hopes that you'll buy their story. While some may not. Look out for other traces of them lying.(Like the points mentioned above)

Know the truth about something before you ask. Or have resources to prove them wrong. Like for instance, my ex husband's girlfriend is a COMPULSIVE LIAR. It's discusting, the trouble that she goes through over a stupid lie.
Monday: She emails me to tell me that she "will" send my daughters insurance card via mail at lunch. After lunch she emails me to tell me that she did send the insurance card at lunch.
Friday: I send her an email, informing her that I haven't received it yet and asked if she had a tracking number. This woman, flips out, calling me every name of the book and telling me that, "she told me that she sent it out certified mail on Monday". I pull up her email and it states absolutely nothing, so I said to her, "well good, at least you have proof of something you did this time".
Tuesday: I finally receive the insurance card in the mail. Yes it's certified, but it wasn't sent out until Saturday. Ohhhh, I couldn't resist.
She is a complusive liar. She lies about everything and anything, just for the state of lying. But I had proof that she lied.

Body language... looking up and to the left (theirs)... minimal eye contact, or eye contact that is too strong.
They may get 'shifty'. Everyone is different... it helps if you know their "non-liar" body language well. I always know when one of my students is lying... his tells are really pronounced. He opens his eyes really wide, and his voice gets deeper and more 'sincere'. His eye contact slides away from my gaze easily. He also fidgets, and taps his feet.
Good luck... and listen for stress in the voice as well... if someone is not prone to saying 'ummh...' a lot, and then they start... kinda interesting... :)

You can usually tell by the faces they make. I know when my brother Eli is lying because he gets this smile on his face... Smirk if you will, When he sees the look on your face.

If you have good instincts...you'll know. Especially if you have known the person for a while. Tell the person of your suspicions and watch how he/she reacts.
