Yes, but depending on what your fueling system is you may have to do some significant changes. Basically you remove the exhaust restrictions so the engine flows more air, but that can result in it running lean if it isn't using a mass flow sensor of some sort. That could burn your exhaust valves, melt your pistons and wreck the engine.
There's also the question of where all that CO laden exhaust is going, headers only means no catalytic converter. If that gets into your breathing air then you are going to get a headache, feel sick, pass out and die... depending how much you get.
It's also going to be loud as hell.
I wouldn't do it.

It won't hurt the engine as long as the exhaust gas doesn't flow over anything such as wiring or painted surface etc.
It will sound like the world is coming to an end.
The engine will actually lose power.

Temporarily, you can do it, just to hear how it sounds. But driving your car that way is probably not legal in most states.
