Sometimes it's quite hard to tell - often the easiest way is to compare kittens.
The females bits are generally closer together than those of the male. Here's some pictures:

Actually this site is very good as it actually has photographs of the difference - rather than just a drawing:

Males carry their penis inside. The easyiest way to tell is a female will have 2 openings really close together(almost touching) and a boy will have a space between the 2 openings. Try looking at them all, if they have litter of mixed sexes (I've seen all female and all male litters before) you should be able to see the difference.

RE:
How do you tell the difference between a boy or girl cat?
My sister's cat just had kittens and my partner and I will be adopting one. My sister's partner said on the phone he couldn't tell the difference between boys and girls. Wouldn't it be much like a dog? Having either a penis or a 'vulva'?
We want a boy so what should we look...

By the time those kittens are old enough to be adopted out, the "equipment" they were born with will be quite evident. Just lift the tail and take a look. In some kittens, the difference is evident sooner, but by the time they are eight weeks old, you will be able to see.

"The sex of kittens is usually easy to determine within the age of approximately six to eight weeks, although it is possible to do so sooner. The male's urethral opening is round, whereas the female's is a slit. Also the distance between anus and urethral opening is greater in males than in females."

Look for the genetals, testicles. They may be small, but should be easy to see at the underside base of the tail if it is a male cat.
