There are a lot of theories as to what causes someone to be "dumb" or unable to speak. In some cases there are physical problems with the vocal chords, (ie they are malformed or underdeveloped), in some cases there are other physical abnormalities that cause a person to be unable to make certain or all sounds.
In other cases, it is a matter of problems with the portions of the brain that control speech. Whether through malformation or though damage these parts of the brain are unable to function properly and therefore the person is unable to make the connection between thought and speech in the brain (in many of these cases there are no other problems it just seems like a short circuit).
Still there are others who cannot speak because they cannot hear. Many deaf people who have been deaf since birth are unable to reproduce speech because they have never heard it and most of our ability to speak is learned through mimicking.
There are no simple answers and there are some cases that are unexplained (ie.. psychosomatic cases where there may be no physical problem but a psychological one that inhibits the ability to speak).

Mostly dumb people are deaf also.
Since they could not hear any sound, they don't even have the idea of producing sound. So they don't try to produce sound.
Nowadays, advanced speech therapy is there to cure them to some extent.

the definition of "dumb" is "unable to speak"
Since they're likely also deaf, they are unable to form inteligent words because they have never heard one. The sounds they make are untelligable, and the politically incorrect call them dumb.

There are various biological reasons people are mute. You can get a lot of information from the Internet. Google Deaf / Mute disorders.

there is a the possibility that the part of the brain that is damaged is also the same area that affects speech, or it's simply that mentally challenged people cannot connect to the part of their brain that is needed to find the correct words or actually use them.

Some people can't talk because they are deaf or blind. Some just didn't go to school or play lots of game. You would want to not be able to talk, right?
