I have been driving a car is correct. If you have only driven one, it would more appropriately be THE car, but A still works. Since you are only able to drive one car at a time, then it is always a car.

Or how about this, unless you have been literally driving a car every second since you were 16, then the absolute correct answer may be, I have driven a car, off and on, since I was 16?

Given our language, a car or cars will be OK.

Do you need to specify that you drive cars? Don't most people drive cars? I would think you might need to specify if you were talking about driving a bus, or a motorcycle, or an 18 wheeler. I would just say I've been driving since I was sixteen.
