It is probably derived from the Red Cross.
Until the 19th century, there were no consensus on markings to identify medics in wat. In 1864 the Geneva Convention was adopted, which included the use of the red cross for this.
The reason why hospitals are usually marked, is probably due to Camillo de Lellis, former soldier, than priest and patron saint of hospitals and nurses. The pope granted him to wear a red cross.
The symbok of First Aid is also a cross, but a green one.

Luke, - an apostle of Jesus was a doctor by profession, he is the one who introduced the plus sign meaning "with the blood of Jesus shed at this CROSS, we can be healed" or literally know "we treat but Jesus heals" Since then, the plus sign (cross) was associated with getting healing.
