Easy. Take I-95 to to the exit for the Holland Tunnel. It should be Route US-1, but there will explicity be a sign for the Holland Tunnel. Pay the toll and you're on the Henry Hudson Parkway in downtown!

From the New Jersey Turnipike:

Take Exit 10 for the Outerbridge Crossing to go into Staten Island, NY
Take Exit 11 for the Garden State Parkway North, to go Upstate.
Take Exit 13 for the Gothels Bridge to go into Staten Island, NY
Exit 14C for the Holland Tunnel for Downtown Manhattan, NY
Exit 16E for the Linclon Tunnel for Mid-Town Manhattan, NY
Exit 18 for the George Washington Bridge for Uptown Manhattan, NY

There are two options:

1. Take exit 14C which will get you to the Holland Tunnel. This is good if you are going to downtown Manhattan or to Brooklyn or even Queens.

2. Take the Turnpike all the way to exit 18, and then continue on the Turnpike (I-95) extension all the way to the George Washington Bridge. This is good if you are going to the Bronx or up into Westchester County.
