We want to do a second coat later and remove the tape then, but are unsure if the tape will still remove like it's suppose to after it has set with paint on it..

It should. Go slowly, try not to get a whole ton of paint on it so you have some clean tape to grab a hold of. I just did an exterior door frame, 3 coats, and tape came off no problemo the next day.

If the paint is dry, you can remove the tape but if you want to put a second coat on, you'll need to replace the tape or risk getting paint where you don't want it. Generally if the first coat was latex paint, it should be dry enough to remove.

Painters tape can stay on for several day, so leave it on and when you put you 2nd coat of paint on, take it off. Painters tape just stays on great and comes of very easily even after a few days.

I always use a razor to cut along the edge of the tape. It gives a real nice clean line. Plus, you avoid those occasions where the tape may pull the paint.
