Greatest negative impact since Christ: The Nicean Council in the 4th century that hijacked the faith and twisted it into the perversion of today's "mainstream" Christianity.

The Apostle Paul had the biggest impact on Christianity in the 1st century after Christ. His writings make up most of the New Testament.

I imagine you don't want us to count the apostle Paul or other apostles either.
The question is difficult. In modern times, in the English speaking world, I could name George Whitefield, the Wesley brothers, Charles Spurgeon, Dwight Moody, or Billy Graham.
However if we go by largest population, I would say Hudson Taylor, as he was used to open up China, and so many folowed after him in other Asian countries. There are tens of millions of believers today in China.
I guess I should have mentioned Luther and Calvin - they were the source of the modern Protestant movements.

Roman Pagan religions.
So much so that the Apostle Paul felt he had worked for nothing as they had gone back to the rituals, observances, and holidays of men. These traditions have been carried over to today, mixing in other beliefs along the way, such as the Yule log, which was used to sacrifice children in northern Europe.

Pope John Paul II . He had a huge impact on the entire Christian and non-Christian world. He was respected by nearly everyone.

This answer is going to vary according to the individual.
Wouldn't it be better to ask who has had the biggest impact on your personal journey with Christ?
For me, it would be men like Lloyd C. Douglas and C. S. Lewis...to name two of the many great writers of Christian fiction.

I believe Saul of Tarsus (Paul) had the biggest impact on Christianity. In fact, all things considered, the religion should be called Paulism, not Christianity.

This is not who I think had the best impact, but who I think had the most impact after Jesus on Christianity.
Paul.
He has basically formed the evangelical church.
In fact, please do not get insulted, his teachings are spoken of a lot more than Jesus' often.
Not to mention he wrote most of the new testement (all of his letters)
He had a major impact on Christianity as we know it today.
