I don't know what 'proof' would be other than health care people accept that there are different frame sizes. At home, you can use the wrist or ankle measurement in relation to height to determine frame size. It's different for men and women. Everyone says I'm "small boned" and I fit into that measurement category. It seems legit.
Women:
height under 5'2"
small = wrist size less than 5.5"
medium = wrist size 5.5" to 5.75"
large = wrist size over 5.75"
height 5'2" to 5' 5"
small = wrist size less than 6"
medium = wrist size 6" to 6.25"
large = wrist size over 6.25"
height over 5' 5"
small = wrist size less than 6.25"
medium = wrist size 6.25" to 6.5"
large = wrist size over 6.5"
Men:
height over 5' 5"
small = wrist size 5.5" to 6.5"
medium = wrist size 6.5" to 7.5"
large = wrist size over 7.5"
I still sometimes weigh more than people who are "bigger" than me even though I look thin, so I guess it isn't an accurate way to explain weighing a lot.

Yes, there is such a thing as big boned. My Doctor informed me that I am big boned and that is why i weigh more on the scale then i look! I was also just informed that if you are German you have a bigger wrist size then other people, I am not sure if this is entirely true but i am German and i have a pretty big wrist. Also i have lost a lot of weight and am only 20 - 30 pounds over weight and i look like i am only 10 pounds over weight and i am also very tall. Also a good determining factor of big boned is if you have ever fallen hard and only sprain body parts with out breaking them you are big boned.

Yes there are medical tests that can be done even simple x-rays can show a big boned tendency. Which is why 2 people can weigh the same but look different and wear different sizes, a persons bone structure can define a lot about them from a physical standpoint. That is why we have giants and dwarfs that exist.

I think the myth part comes in when people use the big boned as an excuse for being fat or overweight. Big boned surely refers to bone structure and not body (here I mean includinng muscle and fat) structure. You can be small boned but by carrying a lot of extra weight seem larger. For instance, my mother is almost a foot shorter than me and her shoulders are are narrower than mine, her hands are narrower and her fingers are shorter. Though she is a little overweight her bones are actually shorter, smaller and narrower. She is not big boned, simply a little overweight. I on the other hand have larger bones in comparison to her. I think the term big boned is used incorrectly when referring to overall body size. It should be used when referring to actual bone size.

I think that is an old Ghetto myth that my people (black people) made up! LMBO... If you watch that show on Vh1 (the one were celebs have to lose weight), bone crusher (rapper) was on there and went to a doctor to see if that myth was true. The doctor said it was false...

I've always heard the expression, but then not too long ago I heard that it's untrue. I can't recall where I heard it but I remember that I believed it, in other words, I trusted the source.
It could just be that I personally find it hard to believe, so I was open to believing it no matter what the source, lol.
