Yes, fatigue can contribute to voice breaking. I myself, currently, is expressing voice breaking because of too much singing yesterday. I have some observed causes of voice breaking below that you might find helpful. Following are some suggestions to prevent voice breaking.

- Dehydration

- Singing too much beyond your vocal range

- Have proper sleeping habits
- Drink lots of water
- Vocalize because singing extensively (like solmization and range scaling)
- Sing with the proper technique
- When you feel tired or you feel unease while singing, STOP! and rest your throat. You might just aggravate your vocal cords and might develop vocal nodes.
- Exercise regularly (it helps in developing endurance of the body expecially if you will incorporate belting in your singing)

when that happens to me, my vocal coach tells me i might be doing something wrong when hitting the notes, like wrong vocal positioning. the correct postion may slightly differ for every singer. what i find generally helpful though is "pushing down". when hitting high notes, don't think high and just let your voice go. instead, when you breathe you should feel your diaphragm or even stomach area "push down and out" for that necessary support when hitting a high note.
another thing that helps is really dropping the jaw, especially when singing at the higher range.
but firstly, she should be sure she's properly vocalized before reaching high notes.
if nothing works, it could just be vocal fatigue, in which case she shouldn't push it and take some vocal rest (i.e. absolutely no talking for a while).

Dehydration can cause some vocal difficulties. It's important to drink lots of water all day if you are a singer. Also, yelling, coughing, & whispering all cause vocal strain. Your sisters troubles could be many things. Maybe she is coming down with a virus or something. Hope it clears up for her.
