Could be a number of things, do you get any kind of error messages?
Your ISP may be blocking the ability to send mail to a different mail server than their own. If you are using a mail server other than your ISPs, change it to your ISP's outgoing SMTP mail server.
You could also test this by click start -> run -> type cmd and hit enter. At teh command prompt type telnet YourOutgoingMailServer 25 (notice there's a space after the mail server name and 25). Get your mail server address by going to tools-> accounts in Outlook, get the smtp address. After typing the command, you'll either get a response from a mail server, something like "EHLO " and some identifier. If you can't connect, then you'll just get a timeout error. If that's the case then you need to use your ISPs mail server, or see if your current e-mail provider provides an alternative port to use for sending mail through.
It could be that your mail server requires authentication to send mail. You'll want to contact them or search their website for a FAQ of some sort. You could turn on authentication by going into accounts in outlook, going to more settings, and making the outbound authentication the same as your incoming authentication (different versions of outlook have different ways of doing this, but if you look around you'll find it).

You could have antivirus that's not working right, that could be blocking it. Try uninstalling your av program, just disabling it may not be enough to test.

Lastly, you could try uninstalling outlook, and re-installing. Just make sure you have your office CD handy.

You need to specify the correct SMTP (Outgoing) email server name under the Server tab in Properties for the email account you are trying to configure.
