OK, all of those neighborhoods are very different, and all have their pros and cons for living there.
Chelsea is a very gay area, so that appeals to some and not others
East Village is a late night, edgier kind of area
Financial District is very busy during the day, but very quiet and not much happening at nights/weekends
Flatiron/Union Square has big loft apartments, can be expensive
Greenwich Village - lots of loud nightlife, historic brownstones, shopping and restaurants, near NYU
Harlem - some people might not feel safe in Harlem, there are lots of low income housing projects on the East Side, but there are also lots of beautiful brownstones and a new and upcoming shopping areas
Inwood/Washington Heights - bigger, older apartments, but quite far from midtown
Lower East Side - used to be an immigrant neighborhood, is now gentrified and lots of new luxury buildings are going up around walk up tenements. Lots of new resturants and bars, some people complain about noise issues. Very trendy area.
Midtown East/West - most business district, harder to find groceries, etc. Crowded and noisy, except on the outer edges
Murray Hill/Kips Bay - kind of boring residential areas, but nice apartments can be found on quiet pretty streets
Soho - expensive! Big loft apartments and expensive boutiques. It used to be a place for artists, but is now way too out of reach
Tribeca - nice family-oriented neighborhood, but also lots of large loft apartments and expensive buildings
Upper East/West Sides - very similar, but the West Side is a little older so the apartments can be a little bigger. Both are expensive close to the park, but you can find bargains going towards the rivers on either side. Lots of shopping, restaurants, families, parks, etc...
I've lived on the Upper East Side for years. I like it because it is very clean and very safe and that is important to me. I don't need to live someplace trendy and cool, I just want a nice apartment at a good price and I have that there.

I live in Herald Sq aka the Garment District, aka Midtown South. Yes, its busy during the day but quiet at night. No bars, restaurant really open. Most people go home. So its quiet and you feel like you own the neighborhood at night. The same can be said of the financial district. It all depends on what you want in your neighborhood. I hate the trendy, nightlife areas like soho, greenwich, west village, meat packing district and chelsea and dont want to live in the snooty Upper East side, so midtown and far downtown are better to suit me.

I like the East Village. Been there 15 years. Still haven't left. Nuf said.

Theres no such thing as "the best place to live" in manhatten come on now but go to the upper east side even though a lot of puerto ricans live there. I used to live there and it felt good to be around my own people..
