Generally an initial screen is done through a blood draw. From there, depending on what's indicated, you could have some sort of internal imaging (CT, MRI, X-RAY). These don't hurt at all. These procedures could be done with or without contrast dyes and whatnot, which may be either drank (none of 'em taste good) or injected via shots or IV.
Also, the bone marrow and spinal fluid might be tested. Both are collected with big needles. Spinal taps (also known as lumbar punctures) aren't too painful because they'll numb the area first. Bone marrow hurts pretty bad for a couple days, but you get over it.
If a tumor or something is found, biopsies are frequently done to determine whether or not it's cancerous.
There ya have it. That's about it.

There are a number of tests and procedures that can detect cancer, and many are specific to the type of cancer suspected. General diagnostic radiographic procedures such as x-ray, including mammogram, and CT scans are commonly used to detect anatomic abnormalities but require biopsy to confirm a diagnosis of cancer. Specific tests include those for cervical cancer, which can be detected by a Pap smear, and prostate cancer screening, which measures PSA levels in the blood. Another growing trend is the use of PET scans, which use radioactive isotopes to show areas of hypermetabolic activity within the body which is consistent with the presence of cancerous cells.
