No bacteria builds up on the bottle, unless you wash the bottle between uses. Rarely would the bacteria be harmful but you can still get sick. If you have access to a lab where you could take cultures from the bottles then you would be able to do the experiment, but other than that I don't know how you would be able to test this as an experiment.

*SIGH*
Only in a throwaway culture...

Ever hear of a canteen? Or a boda bag? Or any of a thousand containers that have been used and reused by mankind since the Ice Age?! Believe it or not, water bottles CAN be reused. For months, and even years at a time!!! Once upon a time, there was this high tech device called a 'bottle brush' that took care of any possible problems from bacteria.

Today's children are truly sad.

Using the same bottle over and over is a great way to reduce waste. Just don't share it with anyone, and make sure to wash it in hot soapy water on a regular basis.

Oh I hope so. I have been refilling the same water bottle and drinking from it for months. Maybe years!
