have you tried to use Yahoo maps or map quest? they're very helpful.
also, look for "land marks" while you're driving. it's easier to remember where you pasted a Wal-Mart on the left, or a Library on the right instead of street names. (You can learn those AFTER you learn your way around.) :)
as for learning to get somewhere new? ride with friends that have lived in the area for years. they always know MANY different ways to get from point A to point B.
and, by all means, do your best to relax. seriously, breathe deep, and your mind will recall the path for you.

I can't tell you one street in my city (pop. 3,000) and i've lived here for 10 years. I swear, i am the worst at getting somewhere or finding someplace. I'm going to invest into a GPS ($80). All you really need to know is the major roads. All you really need is a general idea of the area, but once you stray GPS would be handy to have.

Take a map with you and try joining a taxi training program or something because there is homework for taxi training and you can learn more about new streets and roads!

consider it not getting lose, but exploring new territory. Some of my best trips, local or far, are just getting going in the right direction. If you are going south, just keep following sign, and routes to the south. If going to the south east, follow sign for east and south, you'll always get somewhat close. And you would not believe what new thing you will see. Have a good vacation............ps... we went on vacation once, by just going, and if we saw signs or ads, for different things, we sidetracked to them. What a great time we had.............

Take note of any buildings that you pass and keep a mental picture of what you have passed.As regarding going to somewhere new,every time you get lost, it's somewhere new.

I research where I'm goign and follw driving directions. I often use 1800-555 tell
just say driving directions and is easy just in case you get lost. Try it, its free
