Yes, it is one of the most basic highs there is.
Shapies and other markers and areosol sprays fill the air around them with chemicals that are not air. If you breathe those chemicals rather than air, your brain will not receive enough oxygen and will therefore begin to feel funny.
It is also a very dangerous high for the very same reasons. Once you go too far off the deep end, you won't have enough self awarness to take the sharpy or can away before you die.

Yes they can give you a high...along with brain, liver, and nerve damage. It can kill you the first time you do it, or the 20th time. The mechanism of action for sniffing these chemicals is that it sensitizes your body to the effects of the hormone adrenaline. This hormone normally acts to increase cardiac function, so typically, people who sniff these chemicals die from arrhythmias...in other words, the have a heart attack. Additionally, if you are breathing too much of the chemical and not enough air containing oxygen, you will run into ischemic events which can affect the heart and brain.
All in all...sniffing a sharpie marker to see what it smells like probably won't kill you, but continuous, extended periods of breathing in chemical fumes will lead you into Sudden Sniffing Death Syndrome.
