It really depends on what you and your wife like to do!
You can find pretty much anything that you want here in terms of shopping, theater, art, architecture, sports....
If this is the first and only time you've been here, definitely try and see some of the big sights! One thing that has opened up recently is called "Top of the Rock" and it is an observation deck on the top of Rockefeller Center. It has the best views, much better than the Empire State Building and the lines are not as long.
Another fun thing people might not know about - you can take guided tours of Madison Square Garden that take you back stage and even in the locker rooms of the players! It is fun and doesn't take that long.
Spend some time walking around Central Park, maybe rent a boat on the lake for an hour or so.

Effectively, you have 3 DAYS. Not really a lot of time, so you'll need a really good map. Subway is the quickest and best way to travel (and the cheapest)
Broadway Show (get tix at TKTS in Times Sq.)
St. Patrick's Cathedral / Rockafeller Center - Eat at Top of the Rock.
Visit Greenwich Village (downtown - Bleeker St. etc)
Lincoln Center (b'way/60th st)
Grand Central Terminal/Empire State Bldg./Chrysler Bldg (close enough to walk to all 3)
Statue of Liberty/Ellis Island (might kill a whole day, though)
Take a CIRCLE Line Cruise around Manhattan Island (VERY romantic).
Soooo much to see - So little time.
Have fun!!

Definitely Rockefellar Center. There's the Top of the Rock observation deck, ice-skating, shopping, and the tree! Try to get tickets to the Radio Music Center Christmas show for the complete package. And there's are plenty of places to stop and take a break. A lot of it is underground too.
Pick up tickets to the tour buses, they run everywhere of interest and you can hop on and off. It even goes to the Bronx Zoo.

Eat at Ellen'sStardust Diner 52nd and Broadway. Go up Empire State building and take a Circle cruise around Mahatten (remember to sit on the left of the boat for good views)>

I recommend seeing a Broadway show. or for fun just take the subway to various places like Times Square or 5th ave. The best time to go to the city is at night. Ice Skating at Rockfeller center is fun.Or going to eat out in Hell's Kitchen.

usual tourist places the view of south manhatten looking towards were the twin towers used to be is fantastic, windows of Macy's and Saks 5th Avenue should be dressed up for Christmas they are fantastic and the Rockfellar where they turn the light on the tree is great
