at any given time around the earths skies? At what distance must they remain from each other in their sky-lanes? Vertically and horizontally?

In the U.S. in the En Route environment, the separation standards are 1,000ft vertically, and 5 miles laterally. There are exceptions to this, (i.e. Non-RVSM) but I'd be typing a full length novel to explain it.

Not sure of the exact number of commercial jets in the sky, but if you go to:
http://flightaware.com/
they track the flights in the air throughout U.S. for example they show 4821 flights that they are currently tracking today.
