If you are taking kids GO TO THE TIMES SQUARE TOYSRUS!!!!
They will marvel at the giant lego skyscrapers, the 4 story ferris wheel, and the awesome toy displays. My kids (10 & 8) talked about it for weeks!

Assuming you mean Manhattan, there are two choices:
FAO Schwarz on 5th Avenue between 58th & 59th St., and Toys R Us in Times Square (44th St. and 7th Avenue/Broadway).
FAO Schwarz has a more interesting selection of items, but the Toys R Us is a bit more impressive size-wise and because it has a ferris wheel ($4 per ride) dominating the internal decor.
