After the death of Muhammad, four caliphs, known as the legitimate caliphs by the sunnis, ruled the early muslim empire.
Thes four caliphs were: Abu Bakr, Umar, Utman and Ali. Sunnis recognize them all, but shi'ite only recognize Ali, who was a descendant of Muhammad's uncle, and therefore, part of the prophet's familiy.

The shiites believed that after Muhammad's death the rule should have sticked to his family. And therefore Ali. This conflict between muslims made the rupture.

That's the main reason really, check out Wikipedia if you want to know more.
