Yes, esta with the accent is a verb. It's the third person singular (he, she, it) of "estar", which means to be. In the sentence you provided, you're looking for the third person plural (they), which is estan. This also has an accent on the a. So the correct sentence is "Tus hermanos estan enfermos."

Esta with an accent on the "a" is a verb that means: It/He/She is. The verb in your blank would be plural, and the plural form of esta (with an accent on the "a") is estan (with an accent on the "a")

Yes, está is a verb conjugated in 3rd person, singular.

Estar = To be
yo estoy = I am
tú estás (in Argentina vos estás)= you are
él/ella/eso está = he/she/it is
nosotros estamos = we are
ustedes están (in Spain vosotros estáis) = you are
ellos/ellas/esos están= they are

In your example:
Tus hermanos ESTÁN enfermos.

"está" is different than "esta" or "ésta"

está = x is
esta = this (feminine) eg esta casa= this house
ésta = this one (femenine) eg ésta me gusta más = i like this one better
