Edward Weston's pepper photos, Ansel Adams' "Moon over Hernandez," Garry Winogrand's photograph "New Mexico, 1957" (of a boy in front of a garage), Walker Evans' pictures of street signs, Dorothea Lange's pictures of migrants, Robert Frank's entire book "Americans," Diane Arbus' picture of a child with a grenade, Cindy Sherman's self-portraits, and Robert Maplethorpe's self-portrait with a whip in his ***.

Just off the top of my head, those ones are all pretty key. You might also look at photographs by and of Warhol, some older stuff like Steichen and Stieglitz, Edward Curtiss' dressed-up Native Americans, more Adams contemporaries like Minor White. Other street photographers besides Winogrand and Frank, too, like Lee Friedlander (especially his early work). Of course Stephen Shore should be in the list as well, although as far as I know no single image of his is truly iconic.

Robert Franks book "The Americans" is the quintecitial photography book on life in America in the mid 20th century, a book whose vision is so distilled that 50 years later the book still holds truth. However there were many other great American photographers during that era as well, consider the FSA photographers: Dorothea Lange, Roy Stryker, Walker Evans,Ben Shahn, John Vacahon, Arthur Rothstein and Elmer Ted Johnson.
Of course you cant talk about photography without mention of Ansel Adams whose work in nature conservation and ecology equaled his contributions to photography. You should also look at all the photographers in the F64 group, as Ansel was a member of this group as well. Since we are on the subject of nature conservation the original photographer/conservationist was William Henry Jackson, whose photos of the west helped to establish the US parks system. Alfred Stieglitz should also be mentioned as well, for he was the one responsible not only for bringing modern art to the United States(including Picasso) but also for raising the standards of fine art photography in America as well.
In terms of photographs if you want to talk about 1 single photograph which at one times was considered the most reproduced consider Dorothea Langes, " Migrant Mother". Adams photo, "moonrise over hernandez" is also another great example, but the list really goes on and on. Really though you should take a look at Robert Franks book, The Americans, and there you will find your most distilled an famous photos of American life.

Ditto on the Ansel Adams answer.

The vast majority of space photos are American, so I'd suggest the Apollo pics of the moon, or any number of Hubble pics.
