It would be Mass as this was the first place settled, and as the people came here to be allowed religeous freedom. I would think they set up a church first thing. I doubt they built a church for a while as they were having trouble surviving, but I'm sure they held services and this constitutes establishing a church in my opinion.

There is some evidence (and speculation) that there were some pre-Columbus, Irish monks who wound up on the eastern seaboard and established a monastic community, which would have entailed a church. However, I am not sure with which current state that settlement would correspond.

I would assume those folks on the Mayflower set up a church in Mass as soon as they landed. They did come over for religious beliefs after all.
