The process is called vulcanization, sulfur is added to rubber and heated to stabilize the rubber.there are layers of fabric and steel "belts" for strength. The process varies depending on manufacturer and type of use and specific materials.

The mechanical carcase is wound round a form, starting with a thin rubber liner. A rubber cover is rolled on and then the whole issue is pressed in to a mould and cured under heat and pressure. Very basic version. Wikipedia has a nice article on tires. http://en.wikipedia.org/wiki/Tire
My father used to fit railway tires for part of his working life.

If you have 365 used condoms, you can make a tire from that and call it a GOOD year! Haha, so bad. They are made of rubber, just not THAT type of rubber. The rubber is reinforced with either steel or composite materials such as kevlar or carbon fiber, but steel reinforced rubber is the predominant type.

Modern tyres are constructed of layers, which may include rayon cloth, steel bands and nylon belts all set in a matrix (binder) of rubber.
Hope this helps!
