You should not separate a puppy from its mother until the mother has properly weaned the puppy otherwise you will have problems with the puppy. The standard is 8 weeks, however, you shouldn't simply say, "Okay, 8 weeks is up, let me have the puppy." There are natural processes that the mother must do for the puppy. The most important at that point is weaning.

For many years 49 days (7 weeks) was considered the perfect age to pick up your new puppy.
But,,,,,, here is the catch,,,, you asked at what age should the pup be separated from it's mother, NOT at what age should the pups be separated from each other, and these are two very different things.
Weaning should begin on the 28th day (4 weeks) separating the puppy's from their mother during the day and feeding them three times during the day, and then allowing them to be with their mother at night. At 35 days (5 weeks) they should then be separated from their mother completely,,,,,,,, BUT!!!!! THE LITTER SHOULD REMAIN TOGETHER FOR A MINIMUM OF another 14 DAYS (2 weeks)
The logic for keeping them together for 8 weeks began because of the practice of CERF certifying the puppy's eyes which can not be done before 56 days (8 weeks). Since the practice began many carriers have adapted the rule that a puppy younger than 8 weeks can not be shipped.

About 60 days, or 8 weeks. Pups need to be together and with mom until 8 weeks of age to learn crucial behavior skills.
